Title: Estudo dos mecanismos moleculares da persistência da memória de medo : papel da PKMzeta e dos receptores AMPA contendo a subunidade GluA2

Abstract (native): O envelhecimento se caracteriza pelo declínio das funções orgânicas e cognitivas, e, dentre elas, da memória. Algumas memórias são efêmeras e duram poucos instantes, enquanto outras permanecem enraizadas por toda a vida. Enquanto os mecanismos envolvidos na consolidação da memória já foram mais bem explorados, os processos moleculares responsáveis por sua persistência permanecem incertos. A proteína cinase M ζ (PKMζ) é uma enzima persistentemente ativa, cuja atividade autônoma é responsável pela manutenção da potenciação de longa duração (LTP), um modelo celular de aprendizagem e memória, e também pela persistência de diferentes memórias, através da inibição da endocitose de receptores de glutamato do tipo α-amino-3-hidroxi-metil-5-4-isoxazolpropionato (AMPA) contendo a subunidade GluA2. A memória de medo condicionado ao contexto (MCC) possui grande importância adaptativa para a sobrevivência e, por este motivo, caracteriza-se por seu armazenamento duradouro. Esta forma de aprendizagem recruta diversos circuitos neuronais, dentre eles, o córtex pré-límbico (PrL). No intuito de caracterizar os mecanismos moleculares envolvidos na persistência da memória de MCC, este estudo se propõe a investigar o envolvimento, neste processo, da PKMζ e dos receptores AMPA contendo a subunidade GluA2, bem como sua interação, no PrL, utilizando-se de um modelo experimental de roedores. Para isso, treinamos ratos em um paradigma de MCC e administramos no PrL um inibidor da enzima PKMζ, ZIP (10 nmol/lado), um inibidor da endocitose de receptores AMPA que contêm a subunidade GluA2, GluA23Y (100 pmol/lado) ou o peptídeo inativo GluA23Y(s) (100 pmol/lado), dois ou vinte dias após o condicionamento, e avaliamos a retenção de memória de longa duração vinte e quatro horas depois. Assim, verificamos que as infusões de GluA23Y ou de GluA23Y(s) não afetaram a manutenção das memórias recente ou remota de MCC. Além disso, a inibição da PKMζ no PrL não prejudicou a manutenção da memória recente de MCC. Entretanto, a inibição da PKMζ, vinte dias após o treino, prejudicou a retenção da memória de MCC no teste de memória remota. Este prejuízo foi bloqueado pela administração simultânea de GluA23Y, mas não de GluA23Y(s). Os resultados obtidos sugerem que a PKMζ não é requerida, no PrL, para a manutenção da memória recente de MCC, e que a inibição aguda da endocitose de receptores AMPA contendo a subunidade GluA2 não afeta 9 a manutenção da memória recente nem remota de MCC. Por outro lado, a PKMζ é essencial para garantir a persistência da memória remota de MCC, e ela o faz através do bloqueio da internalização de receptores AMPA contendo a subunidade GluA2 no PrL. Este trabalho confirma o papel crucial do PrL nas memórias de medo e contribui para o entendimento dos mecanismos moleculares responsáveis pela persistência das memórias.
Abstract (english): Ageing is characterized by the decline of organic and cognitive functions, and among them, of memory. While some memories are ephemeral and last only for few moments, others remain deeply rooted during a lifetime. The mechanisms underlying memory consolidation have been relatively well explored. However, the molecular mechanisms underlying the persistence of long-term memory storage remain elusive. Protein kinase M ζ (PKMζ) is a persistently active, and its autonomous activity underlies the maintenance of long-term potentiation (LTP), a cellular model of learning and memory, and also the storage of different types of memory. PKMζ exerts its actions by inhibiting the endocytosis of GluA2-containing α-amino-3-hydroxy-methyl-5-4-isoxazolepropionic acid (AMPA) receptors. Contextual fear conditioning (CFC) memory is a form of learning with high adaptive value due to its importance for survival, and because of that, is characterized by prolonged retention. This memory recruits many neuronal circuits, among them the prelimbic cortex (PrL). In order to characterize the molecular mechanisms underlying CFC memory persistence, this study aims to investigate the involvement, in this process, of PKMζ and GluA2-containing AMPA receptors, as well as their interaction, in the PrL, utilizing a rodent experimental model. For this purpose, we trained rats in a CFC paradigm and administered in the PrL infusions of ZIP (Zeta Inhibitory Peptide, 10 nmol per side), a PKMζ inhibitor, GluA23Y (100 pmol per side), a GluA2-dependent AMPA receptor endocytosis inhibitor or GluA23Y(s), a scrambled control peptide, two or twenty days after conditioning, and evaluated long-term memory retention twenty-four hours later. We verified that infusion of GluA23Y or GluA23Y(s) did not affect the maintenance of recent or remote CFC memory. Besides that, PKMζ inhibition in the PrL did not impair the maintenance of recent CFC memory. However, PKMζ inhibition twenty days after training impaired remote CFC memory retention. This impairment was prevented by the infusion of GluA23Y, but not of GluA23Y(s). These results suggest that PKMζ is not required in the PrL for the maintenance of recent CFC memory. Also, acute inhibition of GluA2-containing AMPA receptor endocytosis does not affect recent or remote CFC memory maintenance. However, PKMζ is essential to ensure the maintenance of remote CFC memory, and it does so by preventing prelimbic GluA2-dependent AMPA receptor internalization. This work confirms a 11 crucial role for PrL in fear memory and also contributes for the understanding of the molecular mechanisms underlying memory persistence.
