dc.description.resumo | A proteína di-hidroneopterina aldolase (DHNA, EC 4.1.2.25) é a terceira enzima da via de biossíntese do folato, catalisando a conversão de 7,8-di-hidroneopterina (DHNP) a 6-hidroximetil-7,8-di-hidropterina (HP) e glicoaldeído (GA). Em Mycobacterium tuberculosis (Mtb), a DHNA é essencial para a sobrevivência do bacilo, caracterizando-se por ser um potencial alvo molecular para o desenvolvimento de fármacos com toxicidade reduzida, visto que esta via está ausente em humanos. Compostos da classe 8-mercaptoguanina foram identificados como inibidores da DHNA e de outras enzimas da via de biossíntese do folato em Staphylococcus aureus e Escherichia coli. No entanto, até o momento, não foram descritos inibidores para a DHNA de Mtb (MtDHNA). Nesse trabalho, uma série de 19 compostos análogos estruturais do 8-mercaptoguanina (8-MG) S-8 substituídos foi sintetizada e avaliada quanto ao potencial inibitório sobre a enzima MtDHNA. Foram identificados 8 compostos com valores de IC50 na faixa de inibição de nanomolar. O mecanismo e as constantes de inibição foram determinados para os 4 compostos com maior potencial inibitório, que apresentaram um perfil de inibição não-competitiva ou competitiva para o substrato DHNP. Além disso, estudos in silico de docagem molecular foram realizados, sugerindo o sítio de interação enzima-inibidor e as conformações dos ligantes. Por fim, os compostos foram avaliados quanto ao potencial de inibição do crescimento da cepa H37Rv de Mtb, e nenhum composto apresentou atividade inibitória sobre o bacilo nas maiores concentrações testadas. | por |
dc.description.abstract | Dihydroneopterin aldolase (DHNA, EC 4.1.2.25) catalyzes the conversion of 7,8-dihydroneopterin (DHNP) to 6-hydroxymethyl-7,8-dihydropterin (HP) and glycolaldehyde (GA) in the folate pathway. DHNA is essential for Mycobacterium tuberculosis (Mtb) bacilli survival and represents an important molecular target for drug development, mainly because this pathway is absent in mammals. 8-mercaptoguanine and its derivatives were identified as inhibitors of DHNA and other enzymes of the folate biosynthesis pathway in Staphylococcus aureus and Escherichia coli. However, no inhibitors have been identified for DHNA from Mtb (MtDHNA). Here, a series of 19 8-mercaptoguanine derivatives S8-functionalized were synthetized and the inhibitory potential against MtDHNA was evaluated. 8 compounds showed IC50 values in the submicromolar range. The mode and inhibition constants were determined for 4 compounds that exhibited the highest enzymatic inhibitory potency, which demonstrated a non-competitive or competitive inhibition profile for DHNP. Further, molecular docking analyses were performed to suggest enzyme-inhibitor interactions and ligand conformations.
