Abstract (english): Introduction: The aims of this study were to assess the in vitro cytotoxicity of orthodontic bands, with and without silver solder, and to compare the viability of the HaCaT, HGF, MRC-5, and Vero cell lines, when exposed to orthodontic band extracts. Material and Methods: An MTT assay was performed on the extracts of the tested materials. The culture medium alone was used as a negative control and copper strip extracts were used as a positive control. Results: A one-way Analysis of Variance (ANOVA) showed a difference between all of the groups in all of the cell lines and Tukey’s post-hoc test revealed no significant differences between the stainless steel orthodontic bands and the negative control group. There was a significant difference between the negative control group and the stainless steel orthodontic bands with silver solder joints in the HaCaT and MRC-5 cell lines. A great variability in the percentages of cell viability was observed. According to these results, the stainless steel orthodontic bands did not induce cytotoxic effects on any of the cell lines. When associated with silver solder, the orthodontic bands decreased the cell viability in the HaCaT, HGF, MRC-5, and Vero cell lines, but there was only a statistical significance in the HaCaT and MRC-5 cell lines. Conclusions: More than one type of cell line must be tested in a cytotoxicity assay due to the outcome variability from one cell line to another.
